Nikon AXR updates boost our upright CLSM capabilities

Laser scanning confocal microscope collected with the Nikon AXR microscope

Our Nikon AXR Upright Confocal Laser Scanning Microscope is being revamped to a more standard focusing-stage format with enhanced automated features and an expanded range of objective magnifications, providing researchers easy access to an array of specialized confocal applications.  The Nikon AXR system enhancements include a new 7-position motored nosepiece configured with a broadened range of objective magnifications, including 10x dry, 20x dry, 40x dry, 50x dry, 25x water immersion, and 60x oil immersion objectives, and a new high-precision motorized, focusing XY stage.  The Nikon AXR confocal module is configured with four high-sensitivity detectors (3 GaAsP + 1 Multi-Alkali PMT) for the separation and detection of standard blue, green, red, and far-red fluorescent signals, such as DAPI, Alexa 488, Alexa 594, Alexa 647.  In addition, the system is configured with a 730nm diode laser for expanded imaging into the near-IR range (e.g., Alexa Fluor 750).  Configured for imaging large fields of view (25mm) with high-speed and high-resolution, the system is optimized for recording fast cellular responses in live tissues and organs.  In addition, a new color camera has been added to the system, providing researchers with the option to directly record conventional fluorescence and transmitted light images, including Brightfield, Phase Contrast, DIC, and Polarized Light techniques.  For improved access to the confocal system, the Nikon AXR was relocated from BPS to the B7 CIPS CAM confocal lab.  For instrument access and training on the Nikon AXR, please contact Dr. Melinda Frame within the Center for Advanced Microscopy.
